No, it’s really just needed for the first time you use them to make sure you don’t burn the wick. After use, even with an empty tank, the wick will still have some juice soaked into it so it’ll be all good without priming.

I’d still advise waiting 5 minutes or so after you fill up before vaping, though. And if you want to prime the coil anyway it won’t do any harm 🙂
